Using orange dyes to perform multi-color cytometry with the 405 nm laser could be a stretch in the past. Existing fluorochromes could lack brightness and/or have extended spill-overs into adjacent channels, thereby reducing your assay sensitivity. Krome OrangeTM is the new orange dye to overcome these hurdles. In contrast to other dyes available on the market, Krome OrangeTM is maximally excited with the violet laser (405nm), whereas excitation with a 488 nm laser is close to zero, thus resulting in almost no inter-laser overspill into FITC. Furthermore, the emission peak of Krome OrangeTM (530 nm) is far away from Pacific BlueTM (450 nm), thus leading to only very limited intra-laser compensation. Because of this limited inter- and intra- laser spill overs, Krome OrangeTM is the NEW reference orange dye for 405nm excitation in the market to offer you additional flexibility for your violet assays without compromising sensitivity.
